Antibody Immunogen Description: Corresponds to amino acid residues W595 to L602. The effect of Y599 phosphorylation is unclear. This is one of the major in vivo phosphorylation sites in EphA1 with 34 mass spectrometry reports recorded in PhosphoSitePlus.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: EphA1 is a 108-130 kDa protein-tyrosine kinase of the TK group and Eph family. It is activated by binding of ephrin-A1 (EFNA1)on a presenting cell, which is likely to induce dimerization and autophosphorylation. This kinase is moderate to highly expressed in most tested human tissues, and located in the plasma membrane, cytoplasm, and sometimes in the nucleus. Orthologues of EphA1 are highly conserved in mammals and birds. EphA1 has been implicated in mediating embryogenesis and early developmental, particularly in the nervous system. It regulates angiogenesis, cell attachment, and inhibits motility. EphA1 seems to be a marker of the differentiated normal epidermis, and its downregulation in nonmelanoma skin cancer may contribute to carcinogenesis of these very frequent human tumours. EphA1 represents a new potential prognostic marker and therapeutic target in nonmelanoma skin cancer. EphA1 has also been linked with the development of glioblastomas, head and neck cancer and breast carcinomas (pleomorphic lobular).
