Antibody Immunogen Description: Corresponds to amino acid residues E776 to R782. Y779 phosphorylation stimulates phosphotransferase activity. Phosphorylation regulates cell adhesion, cytoskeletal reorganization, and induces molecular association with CRK. This phosphosite is located in the kinase activation loop between catalytic subdomains VII and VIII. This is the major in vivo phosphorylation site in EphA3 (≥723 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.This antibody was also subject to negative purification over phosphotyrosine-agarose.

Scientific Background: EphA3 is a receptor protein-tyrosine kinase of the TK group and Eph family. It features an extracellular domain (with Cys-rich region and 2 fibronectin type III repeats), a single transmembrane domain, and a cytoplasmic protein-tyrosine kinase domain. It is activated by binding ephrin-A1, A2, A3, A4 or A5 (with high affinity for EFNA5), and subsequent receptor dimerization. It mediates contact-dependent cell signalling, primarily in nervous system development and tissue patterning. It regulates cell migration and adhesion by binding membrane-bound ephrin-A ligands, often promoting cell repulsion. Phosphorylation of Tyr-596, Tyr-602 and Tyr-779 increases phosphotransferase activity and interaction with Crk. It is widely expressed in human tissues. Upregulation of EphA3 is common in glioblastomas, lung cancer and melanomas. Its mutation pattern in human tumours is very characteristic of a tumour suppressor protein. Somatic mutations in EPHA3 are found in breast, lung, and pancreatic cancers, often leading to reduced receptor function. EphA3 function is inhibited and EFNA5 binding interrupted with the V133E, and F152L mutations. EphA3 is linked to Pleomorphic Rhabdomyosarcoma, which is a rare disease which is related to colorectal cancer, and can also affect the uterus, heart, and skeletal muscle tissues.
