Antibody Immunogen Description: Corresponds to amino acid residues M591 to P597; in the region between the transmembrane domain and the kinase catalytic domain. One of the two major in vivo phosphorylation sites in EphB1.Y594 phosphorylation induces binding to Nck1. This is one of the two major in vivo phosphorylation sites in EphB1 (≥132 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). EphB1 is known to be phosphorylated at this site in vitro by EphB1.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: EphB1 is a receptor protein-tyrosine kinase of the TK group and Eph family. It feature an extracellular ligand-binding region (with an immunoglobulin-like domain, a cysteine-rich region, two fibronectin type-III domains), a transmembrane domain, and a cytoplasmic protein-tyrosine kinase domain, followed by a SAM (sterile alpha motif) domain). It mediates cell contact signalling via binding Ephrin B family (EFNB1, EFNB2 and EFNB3) of ligands. Phosphorylation at Tyr-594 induces interaction with Nck1 and the SH2 domain of Grb10. Phosphorylation at Tyr-928 induces interaction with Grb2 and Grb7. Interaction with Shc1 and Src involves Tyr-600 phosphorylation, and Shc1 binding also occurs with Tyr-778 phosphorylation. EphB1 is primarily expressed in developing neural structures (brain, hippocampus) and also found in vascular epithelium and the kidney. EphB1 controls neuronal cell development through mediating axon guidance. It is crucial for retinal axon guidance at the optic chiasm, synaptic plasticity, and vascular development. It also controls chemotaxis, proliferation, and polarity of hippocampal neuronal progenitor cells. The SH2 domain of GRB10 cannot bind EphB1 with an Y928F mutation. EphB1 has been linked with the development of gastric adenocarcinomas and ovarian undifferentiated carcinoma. Loss of EphB1 is associated with metastasis in serous ovarian cancer.
