Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: The SARS-CoV-2 virus spike protein (S) attaches the virion to the cell membrane by interacting with primarily the host receptor angiotensin-converting enzyme-2, initiating the infection. During the infection, the S protein is cleaved into two subunit. This cleavage can be catalyzed by TMPRSS2 or during endocytosis by host CSTL. When S2/S2' has been cleaved, binding to the receptor triggers direct fusion at the cell membrane. When S2/S2' has not been cleaved, binding to the receptor results in internalization of the virus by endocytosis using host TFRC and GRM2 and leading to fusion of the virion membrane with the host endosomal membrane. Alternatively, S can use the NRP1/NRP2, integrin and ASGR1 as entry receptors. The use of NRP1/NRP2 receptors by the S protein for host cell entry may explain the infection of human olfactory epithelial cells, which express these proteins at high levels but ACE2 at low levels.
