Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: The SARS-CoV-2 virus Non-structural protein-15 (NSP15) gene encodes a uridylate-specific endoribonuclease that plays a role in viral transcription/replication and prevention of the simultaneous activation of host cell dsRNA sensors, such as MDA5/IFIH1, OAS, and PKR. It likely acts by degrading the 5'-polyuridines generated during replication of the poly(A) region of viral genomic and subgenomic RNAs. It catalyzes a two-step reaction in which a 2'3'-cyclic phosphate (2'3'-cP) is first generated by 2'-O transesterification, which is then hydrolyzed to a 3'-phosphate (3'-P). If not degraded, poly(U) RNA would likely hybridize with poly(A) RNA tails and activate host dsRNA sensors. It may bind genomic dsRNA in association with the replication-transcription complex (RTC), and play a role in nsp12 discontinous transcription.
