Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: The SARS-CoV-2 virus Non-structural protein-3 (NSP3) gene encodes a papain-like protease that is responsible for the cleavages located at the N-terminus of the replicase polyprotein. It participates together with NSP4 in the assembly of virally-induced cytoplasmic double-membrane vesicles that are necessary for viral replication. It antagonizes innate immune induction of type I interferon by blocking the phosphorylation, dimerization and subsequent nuclear translocation of host IRF3. It also likely prevents host NF-kappa-B signaling. In addition, PL-PRO possesses a deubiquitinating/deISGylating activity and processes both Lys-48- and Lys-63-linked polyubiquitin chains from cellular substrates. It preferentially cleaves ISG15 from antiviral protein IFIH1 (MDA5), but not RIGI. It can facilitate host ADP-ribosylation by ADP-ribose. It plays a role in the formation and maintenance of double membrane vesicles (DMVs) replication organelles.
