This phosphopeptide may also be useful as a substrate for screening the phosphatase activity of protein phosphatases. The peptide sequence is located in the region between the C1_1 and kinase catalytic domains. This is the major in vivo phosphorylation site in Raf1. Phosphorylation inhibits phosphotransferase activity, reduces H-Ras-1 and PPP2CB binding, and induces binding of 14-3-3-beta and zeta. It appears to necessary for PKC-alpha activation of Raf1.
