Antibody Phosphosite: Y580
Protein UniProt: Q14738
Protein SigNET: PPP2R5D
Antibody Type: Polyclonal
Antibody Host Species: Rabbit
Antibody Immunogen Source: Synthetic phosphopeptide patterned after human PPP2R5D
Antibody Immunogen Sequence: PQDV(pY)TIKAC
Antibody Immunogen Description: Corresponds to amino acid residues P576 to A584. The effect of phosphorylation of Y580 is unclear, but it is a major site of phosphorylation (≥95 reports by mass spectrometry in PhosphoSitePlus).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.
Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.
Antibody Concentration: 0.5 mg/ml
Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol
Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.
Product Use: Western blotting | Antibody microarray
Antibody Dilution Recommended: 2 µg/ml for immunoblotting
Antibody Species Reactivity: This antibody detects the target phosphoprotein in the following species due to conservation of amino acid sequence: Human | Chimpanzee | Rhesus macaque | Dog | Rat | Platypus | Zebra fish | Honey bee.

Scientific Background: PPP2R5D (Serine/threonine-protein phosphatase 2A 56 kDa regulatory subunit delta isoform) belongs to the phosphatase 2A regulatory subunit B56 family, Isoform Delta-2. The regulatory B56-delta subunit of the PP2A enzyme modulates substrate selectivity and catalytic activity, and also might direct the localization of the catalytic enzyme to a particular subcellular compartment. It is a 70 kDa protein that binds to PP2A scaffold (A) and catalytic (C) subunits in a trimeric complex. It helps to facilitate dephosphorylation of specific protein targets to control signaling pathways (e.g., AKT-mTOR). It is widely expressed, with the Delta-1 isoform highly expressed in the brain, with intermediate-to-high levels in the striatum and thalamus, and lower levels in the cortex. Mutations causing neurodevelopmental disorders, including intellectual disability, developmental delays, macrocephaly, autism, epilepsy (including complex partial and multifocal), hypotonia (weak muscle tone), and autism spectrum disorder. Pathogenic variants usually change conserved amino acids (often converting a negative glutamic acid to a positive lysine), which disrupts the binding of the PP2A subunits and impairs the enzyme's ability to properly dephosphorylate targets, leading to increased activity in specific signaling pathways.
