Target Full Name: Serine/threonine-protein phosphatase 2A catalytic subunit alpha and beta isoforms

Target Alias: EC 3.1.3.16; PP2AA; PP2A-alpha; PP2Calpha; Protein phosphatase 2 (formerly 2A) catalytic subunit, alpha isoform; Replication protein C; RP-C; Serine/threonine protein phosphatase 2A, catalytic subunit, alpha isoform

Product Type Specific: PPP2CA protein phosphatase phosphosite-specific antibody

Antibody Code: PP549

Antibody Target Type: Phosphosite-specific

Antibody Phosphosite: Y284

Protein UniProt: P67775

Protein SigNET: PPP2CA

Antibody Type: Polyclonal

Antibody Host Species: Rabbit

Antibody Immunogen Source: Synthetic phosphopeptide patterned after human PPP2CA

Antibody Immunogen Sequence: DTLK(pY)SFL(βA)C

Antibody Immunogen Description: Corresponds to amino acid residues D280 to L287. The effect of Y284 phosphorylation is unclear. This is the major in vivo phosphorylation site in PPP2CA based on ≥19 mass spectrometry reports recorded in PhosphoSitePlus. PPP2CA is known to be phosphorylated at this site in vitro by Fyn, and Src.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.

Antibody Concentration: 0.5 mg/ml

Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol

Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.

Product Use: Western blotting | Antibody microarray

Antibody Dilution Recommended: 2 µg/ml for immunoblotting

Antibody Species Reactivity: This antibody detects the target phosphoprotein in the following species due to conservation of amino acid sequence: Human | Chimpanzee | Dog | Rat | Mouse | Platypus | Chicken | Honey bee | Sea urchin | Red bread mold.

Scientific Background: PPP2CA (Serine/threonine-protein phosphatase 2A catalytic subunit alpha isoform) belongs to the PPP phosphatase family, PP-1 subfamily. It serves as the catalytic component of the PP2A heterotrimeric enzyme. It forms a catalytic core with a 65 kDa scaffolding (A) subunit, which then associates with various regulatory (B) subunits to define substrate specificity. PPP2CA is involved in microtubule regulation, mitosis, and autophagy, with its activity directed by diverse regulatory subunits. It acts as a critical tumour suppressor and regulator of cell growth, division, and signaling by dephosphorylating targets like p53/TP53, c-Myc, casein kinase 2, mitogen-stimulated S6 kinase, and AKT. It activates RAF1 by dephosphorylating it at its inhibitory Ser-259 phosphosite. It is a major phosphatase for microtubule-associated proteins (MAPs). PP2A also modulates the activity of phosphorylase B kinase and MAP-2 kinase. It cooperates with SGO2 to protect centromeric cohesin from separase-mediated cleavage in oocytes specifically during meiosis I. It is ubiquitously expressed throughout the cell, including the cytoplasm and nucleus. It acts as a tumour suppressor by inhibiting oncogenic pathways such as RAF1, AKT, and WEE1. Mutations or dysfunction can lead to cancer and neurodevelopmental disorders, such as Houge-Janssens syndrome-3 This description may include information annotated by UniProt and/or Google AI.
