Antibody Immunogen Description: Corresponds to amino acid residues R31 to T38. T35 phosphorylation stimulates phosphatase activity through inhibition of this inhibitor. This is a minor in vivo phosphorylation site in PPP1R1A based on ≥4 mass spectrometry reports recorded in PhosphoSitePlus. PPP1R1A is known to be phosphorylated at this site in vitro by PKACa (PRKACA).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: PPP1R1A (Protein Phosphatase 1 Regulatory Subunit 1A; Inhibitor-1) is a 20-28 kDa cytoplasmic protein that inhibits Protein Phosphatase 1 (PP1), and regulates cell signaling, glycogen metabolism, and cardiac contractility. Its inhibitory activity towards PP1 catalytic activity is increased by cAMP-mediated phosphorylation at Thr-35 and decreased by the protein phosphatase calcineurin (PP2B). It does not inhibit type-2 phosphatases. It is highly expressed in tissues like the heart and pancreas. It is implicated in Ewing sarcoma, where it is upregulated to promote tumour growth.
