Antibody Immunogen Description: Corresponds to amino acid residues R430 to S437. The effect of Y434 phosphorylation is unclear. This is a major in vivo phosphorylation site in PPP1R16A based on ≥13 mass spectrometry reports recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: PPP1R16A (Protein phosphatase 1 regulatory subunit 16A; MYPT3) is a membrane-bound regulatory subunit for Protein Phosphatase 1 (PP1), which is involved in the regulation of the phosphorylation state of myosin, which is essential for intracellular movement and muscle contraction. It directs the PP1 catalytic subunit to target substrates, such as myosin light chain, and acts as an inhibitor of PP1 activity. It features five ankyrin repeats, a PP1-binding site, a potential Walker A ATP/GTP-binding site, and a C-terminal CAAX box for farnesylation. While many MYPT members inhibit PP1 when phosphorylated, phosphorylation of PPP1R16A by Protein Kinase A (PKA) has been shown to activate the catalytic activity of the PP1 complex.
