Antibody Immunogen Description: Corresponds to amino acid residues R493 to P499. The effect of MYPT1 Y496 phosphorylation is unclear. This is a major in vivo phosphorylation site in MYPT1 based on ≥439 mass spectrometry reports recorded in PhosphoSitePlus. PPP1R12A is known to be phosphorylated at this site in vitro by ILK.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: PPP1R12A (Protein phosphatase 1 regulatory subunit 12A; MYPT1) is a 115 kDa protein that acts as the regulatory targeting subunit of myosin light chain phosphatase (MLCP) with the catalytic subunit (PP1cδ) to modulate phosphatase activity. It crucially regulates vascular tone, smooth muscle contraction, cell migration, and cytoskeletal dynamics by controlling myosin light chain phosphorylation. It mediates binding to myosin. As part of a PPP1C complex, it involved in dephosphorylation of PLK1. It is capable of inhibiting HIF1AN-dependent suppression of HIF1A activity. Rho-associated kinase (ROCK) inactivates the myosin phosphatase complex by phosphorylating PPP1R12A, leading to increased myosin light chain (MLC) phosphorylation and contraction.
