Antibody Immunogen Description: Corresponds to amino acid residues A302 to L309. The effect of Y306 phosphorylation is unclear. This is a major in vivo phosphorylation site in PPP1CB based on ≥314 mass spectrometry reports recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: PPP1CB (Protein Phosphatase 1 Catalytic Subunit Beta) is a protein-serine/threonine phosphatase that belongs to the PPP phosphatase family, PP-1 subfamily. PPP1CB is sometimes referred to as PP1-delta (PP1\(\delta \)) and is one of three catalytic subunits (alpha, beta and gamma) of PP1. As a broad specificity phosphatase, it regulates diverse processes like cell division, metabolism including glycogen synthesis and breakdown, muscle contraction, transcription and synaptic plasticity. It exists as a ~37 kDa catalytic subunit, widely expressed in tissues, and forms specific complexes with >200 regulatory proteins to control hundreds of targets. It is a component of the PTW/PP1 phosphatase complex, which functions in the control of chromatin structure and cell cycle progression during the transition from mitosis into interphase. It regulates NEK2 function in terms of its kinase activity and centrosome number and splitting, both in the presence and absence of radiation-induced DNA damage.
