Antibody Immunogen Description: Corresponds to amino acid residues G139 to D146. The effect of S143 phosphorylation is unclear. This is the major in vivo phosphorylation site in HePTP (≥99 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). This human phosphosite is highly conserved in mammals and also found in the chicken.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: HePTP (haematopoietic protein tyrosine phosphatase; tyrosine-protein phosphatase non-receptor type 7; PTPN7) belongs to a subgroup of phosphatases that include STEP and PTP-SL, which regulate MAPKs in different tissues. It is a 38 kDa non-receptor protein-tyrosine phosphatase that is primarily expressed in T-cells, B-cells, and hematopoietic tissues such as thymus and spleen. It consists of a C-terminal catalytic domain (residues 44-339) and an N-terminal extension (residues 1-43), which features a Kinase Interaction Motif (KIM). The KIM enables high-affinity binding to ERK1/2 and p38 MAP kinases, and allows HePTP to dephosphorylate the activating phospho-Tyr-185 in their activation loops. HePTP activity is regulated by PKA-mediated phosphorylation at Ser-23, which causes the phosphatase to dissociate from ERK2/p38, allowing the MAPKs to translocate to the nucleus.
