Antibody Immunogen Description: Corresponds to amino acid residues F1213 to L1219. Y1216 phosphorylation stimulates phosphatase activity. Phosphorylation at this site has only be reported in a single mass spectrometry study. This human phosphosite is highly conserved in vertebrates and also found in fish. CD45 is known to be phosphorylated at this site in vitro by CSK.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: CD45 (Receptor-type tyrosine-protein phosphatase C; PTPRC) is a 180-220 kDa receptor-tyrosine protein phosphatase that belongs to the Receptor class 1/6 subfamily, and is present on the surface of almost all hematopoietic cells. It is not found on mature erythrocytes or plasma cells. It is required for T-cell activation through the antigen receptor. It functions as a positive regulator of T-cell coactivation upon binding to DPP4. It has a heavily glycosylated, highly variable extracellular domain, a transmembrane domain, and a cytoplasmic tail with two highly conserved tyrosine phosphatase domains. The first PTPase domain has main enzymatic activity, while the second one influences the substrate specificity of the first one. Upon T-cell activation, it recruits and dephosphorylates SKAP1 and FYN. Its dephosphorylation of LYN, activates its phosphotransferase activity. It acts as a receptor for human cytomegalovirus protein UL11 and mediates binding of UL11 to T-cells, leading to reduced induction of tyrosine phosphorylation of multiple signalling proteins upon T-cell receptor stimulation and impaired T-cell proliferation. Through alternative splicing of exons 4, 5, and 6 (A, B, C), different isoforms are created (e.g., CD45RA, CD45RO) that change based on cell type and activation state. Eight human isoforms are described. Isoforms 1, 2, 3, 5, and 7, whereas isoforms 4, 6 and 8 are detected in thymocytes.
