Product Type Specific: PTPRA protein phosphatase phosphosite-specific antibody

Antibody Code: PP521

Antibody Target Type: Phosphosite-specific

Antibody Phosphosite: Y798

Protein UniProt: P18433

Protein SigNET: PTPRA

Antibody Type: Polyclonal

Antibody Host Species: Rabbit

Antibody Immunogen Source: Synthetic phosphopeptide patterned after human PTPRA

Antibody Immunogen Sequence: CAFSD(pY)ANF

Antibody Immunogen Description: Corresponds to amino acid residues A794 to F801. Y798 phosphorylation stimulates phosphatase activity and regulates cytoskeletal reorganization. This is a major site of in vivo phosphorylation of PTPRA (≥2407 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). PTPRA is known to be phosphorylated at this site in vitro by Src.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.

Antibody Concentration: 0.8 mg/ml

Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol

Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.

Scientific Background: PTPRA (Protein Tyrosine Phosphatase Receptor Type A) belongs to the protein-tyrosine phosphatase family, Receptor class 4 subfamily. It regulates cell growth, differentiation, and adhesion by removing phosphate groups from tyrosine residues on target proteins. As a receptor-type PTP, it is ubiquitously expressed, crucial for activating Src family kinases, and implicated in progression and neuronal migration. Its functions include dephosphorylating and activating Src family kinases, regulating integrin-mediated signaling, focal adhesion assembly, and modulating synaptic transmission. It features an extracellular domain, a single transmembrane segment, and two tandem intracytoplasmic catalytic domains. It is found in the plasma membrane and focal adhesions. Elevated expression is linked to tumour progression in various cancers, including breast, colon, and lung cancer. It acts as a regulator in RET signaling for medullary thyroid cancer and is involved in microbial pathogenesis, such as the action of Helicobacter pylori VacA toxin.
