Antibody Immunogen Description: Corresponds to amino acid residues L483 to E489. The effect of S486 phosphorylation is unclear. This is the major site of in vivo phosphorylation of PTPN14 (≥55 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). This human phosphosite is highly conserved in vertebrates and also found in the frog. Note that the PTPN14 S486 site has the same sequence as S492 in PTPN21 (PTPD1; Tyrosine-protein phosphatase non-receptor type 21).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: PTPN14 (Tyrosine-protein phosphatase non-receptor type 14) is a cytoplasmic tyrosine-protein phosphatase that belongs to the protein-tyrosine phosphatase family, Non-receptor class subfamily. It plays a role in the regulation of lymphangiogenesis, cell-cell adhesion, cell-matrix adhesion, cell migration, cell growth, and angiogenesis, modulates cellular motility and cytoskeleton-associated processes, and also regulates TGF-beta gene expression, thereby mediating the epithelial-mesenchymal transition. It features an N-terminal FERM domain for membrane/cytoskeleton association and a C-terminal catalytic PTP domain. It dephosphorylates targets like beta-catenin at adherence junctions and p130Cas to regulate cell adhesion and migration.
