Antibody Immunogen Description: Corresponds to amino acid residues R36 to Y42. S39 phosphorylation inhibits phosphatase activity. This is one of the minor sites of in vivo phosphorylation of PTPN12 (≥15 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). This particular human phosphosite is highly conserved in vertebrates and also found in the sea urchin. PTPN12 is known to be phosphorylated at this site in vitro by PKACa (PRKACA), and PKCa (PRKCA).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: PTPN12 (Protein tyrosine phosphatase non-receptor type 12; PTP-PEST) is a ubiquitously expressed cytosolic phosphatase that belongs to the protein-tyrosine phosphatase family, Non-receptor class 4 subfamily. It regulates cell growth, cytoskeletal structure and adhesion, differentiation, and motility by dephosphorylating targets such as ErbB2 (at Tyr-1112, Tyr-1196 and Tyr-1248), c-ABL, FAK, and p130Cas. It is involved in T-cell regulation, specifically in secondary responses and promoting the formation of cell homoaggregates. It possesses an N-terminal PTP catalytic domain and a C-terminal PEST motif that mediates protein-protein interactions. As a tumour suppressor, it inhibits oncogenic transformation. It acts as a negative regulator of tyrosine kinases by removing phosphate groups, influencing cell migration, adhesion, and proliferation. PTPN12 has been found to suppress the progression of cancers such as hepatocellular carcinoma and osteosarcoma by targeting receptor tyrosine kinases. Reduced PTPN12 expression is associated with cancer metastasis and poor prognosis in various cancers, including non-small cell lung cancer and nasopharyngeal carcinoma.
