Target Full Name: Protein phosphatase 1 regulatory subunit 12B

Target Alias: Myosin phosphatase regulatory subunit; Myosin phosphatase target 2; Myosin phosphatase-targeting 2; Myosin phosphatase-targeting subunit 2; MYPT2

Product Type Specific: PPP1R12B protein phosphatase phosphosite-specific antibody

Antibody Code: PP503

Antibody Target Type: Phosphosite-specific

Antibody Phosphosite: T646

Protein UniProt: O60237

Protein SigNET: PPP1R12B

Antibody Type: Polyclonal

Antibody Host Species: Rabbit

Antibody Immunogen Source: Synthetic phosphopeptide patterned after human PPP1R12B

Antibody Immunogen Sequence: RRS(pT)QGV(βA)C

Antibody Immunogen Description: Corresponds to amino acid residues R643 to V649. T646 phosphorylation inhibits activity. This is a minor site of in vivo phosphorylation of PPPP1R12B (≥9 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). This human phosphosite is conserved in mammals and found in the rat. PPP1R12B is known to be phosphorylated at this site in vitro by ROCK1 (ROK-beta).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.

Antibody Concentration: 1 mg/ml

Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol

Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.

Product Use: Western blotting | Antibody microarray

Antibody Dilution Recommended: 2 µg/ml for immunoblotting

Antibody Species Reactivity: This antibody detects the target phosphoprotein in the following species due to conservation of amino acid sequence: Human | Chimpanzee | Rhesus macaque | Dog | Rat | Mouse.

Scientific Background: PPP1R12B (Protein phosphatase 1 regulatory subunit 12B; MYPT2) is a 115 kDa protein that acts as the regulatory targeting subunit that binds with the catalytic subunit (PP1cδ) to modulate muscle contactility. It regulates myosin phosphatase activity and augments Ca2+ sensitivity of the muscle contractile apparatus. Rho-associated kinase (ROCK) inactivates the myosin phosphatase complex by phosphorylating PPP1R12B at Thr-646, leading to increased myosin light chain (MLC) phosphorylation and contraction. It is primarily expressed in skeletal muscle, heart, and brain, with isoforms 3 and 4 being heart-specific. It has been associated with various conditions including abdominal aortic aneurysm, colorectal cancer, and potential roles in insulin signaling. It has been identified as a target for insulin-stimulated phosphorylation. This description may include information annotated by UniProt and/or Google AI.
