Antibody Immunogen Description: Corresponds to amino acid residues S309 to H316. The effect of SYT1 Y312 phosphorylation is unclear. This is a major phosphorylation site in SYT1 based on ≥43 mass spectrometry reports recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: SYT1 (Synaptotagmin-1) is an essential Ca2+-sensing, integral membrane protein of synaptic vesicles that belongs to the synaptotagmin family. Upon Ca2+ influx, it mediates fast exocytosis (neurotransmitter release) through regulation of membrane interactions for trafficking of synaptic vesicles at the active zone of the synapse. It triggers synchronized neurotransmitter release by mediating Ca2+-dependent vesicle docking and fusion with the SNARE complex. It features a luminal domain, a transmembrane domain, and two cytosolic C2 domains (C2A and C2B). The C2A and C2B domains bind Ca2+ and anionic phospholipids, which require the presence of both an acidic head group and a diacylglycerol backbone. A Ca2+-dependent interaction between synaptotagmin and putative receptors for activated protein kinase C (PKC) also exists. It can bind to at least three additional proteins in a Ca2+-independent manner; these are neurexins, syntaxin and AP2. Plays a role in dendrite formation by melanocytes, where it is expressed. Mutations in the SYT1 gene are linked to a severe neurodevelopmental syndrome, characterized by developmental delay, involuntary movements, and hypotonia.
