Antibody Immunogen Description: Corresponds to amino acid residues S1080 to A1088. The effect of SYNJ1 S1084 phosphorylation is unclear. This is a major site of in vivo phosphorylation of SYNJ1 based on ≥14 mass spectrometry reports recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: SYNJ1 (Synaptojanin) is a critical phosphoinositide phosphatase that is highly enriched in the brain. It catalyzes the breaks down PI(4,5)P2 to PI(4)P or phosphatidylinositol. By regulating membrane PI(4,5)P2 levels, it is essential for synaptic vesicle endocytosis (recycling) and endocytosis and clathrin-mediated endocytosis, allowing vesicles to be recycled for further neurotransmitter release. It also plays a role in autophagosome biogenesis. It is composed of three main domains: an N-terminal Sac1-like inositol domain, a central 5′-phosphatase domain (5′PP), and a C-terminal region that binds to endocytic proteins like clathrin. The two phosphatase domains (Sac1-like and 5-phosphatase) dephosphorylate lipids. SYNJ1 is located in the presynaptic terminal, attached to synaptic vesicles and endocytic intermediates.
