Antibody Immunogen Description: Corresponds to amino acid residues S569 to K577. Phosphorylation of T574 inhibits exocytosis. This has not been identified as an in vivo phosphorylation site of STX1B based mass spectrometry reports recorded in PhosphoSitePlus. STXBP1 is known to be phosphorylated at this site in vitro by CDK5, ERK1 (MAPK3), and ERK2 (MAPK1).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: STXBP1 (Syntaxin-binding protein 1; Munch18-1) is a regulatory chaperone protein that belongs to the STXBP/unc-18/SEC1 family. It participates in the regulation of synaptic vesicle docking and fusion through interaction with GTP-binding proteins. It is essential for neurotransmission and binds syntaxin, a component of the synaptic vesicle fusion machinery in the SNARE complex probably in a 1:1 ratio. It can interact with syntaxins 1, 2, and 3, but not with syntaxin 4. It is an arch-shaped, 68-kDa protein that forms a cavity to bind the Habc domain and the SNARE motif of syntaxin-1. It may serve a role in determining the specificity of intracellular fusion reactions. It is highly expressed in brain and spinal cord, and particularly enriched in axons. Pathogenic mutations causing haploinsufficiency—where not enough functional protein is produced—lead to intellectual disability, developmental delays, and STXBP1-related disorders such as STXBP1-RD), which include infantile epileptic encephalopathy-4 (also known as Ohtahara syndrome).
