Antibody Immunogen Description: Corresponds to amino acid residues E183 to T190. S187 phosphorylation stimulates its activity. This is a minor in vivo sitesfor phosphorylation of SNAP25 based on ≥1 mass spectrometry reports recorded in PhosphoSitePlus. SNAP25 is known to be phosphorylated at this site in vitro by PKACa (PRKACA), PKCa (PRKCA), and ROCK2 (ROK-alpha).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: SNAP25 (Synaptosomal-associated protein 25) is a 25 kDa membrane bound protein anchored to the cytosolic face of membrane via palmitoyl side chains in the middle of the molecule (1). It functions in the regulation of neurotransmitter release, acting as a key component of the SNARE complex to facilitate synaptic vesicle fusion at the presynaptic plasma membrane. It is proposed to account for the specificity of membrane fusion and to directly execute fusion by forming a tight complex that brings the synaptic vesicle and plasma membranes together (2). It forms a core four alpha-helix bundle with syntaxin-1 and synaptobrevin (VAMP) to drive the docking and fusion of vesicles with the membrane. SNAP25 exists as two alternatively spliced isoforms, SMAP25A and SNAP25B. These isoforms are differentially expressed in neurons and neuroendocrine cells and they differ by 9 amino acids in the central portion of the protein (3). SNAP25 is anchored to the cytoplasmic face of the plasma membrane via palmitoylated cysteine residues, rather than a transmembrane domain. Mutations or imbalances in SNAP25 are associated with epilepsy, intellectual disability (SNAP25-DEE), and neuropsychiatric disorders like ADHD.
