Antibody Phosphosite: T478
Protein UniProt: P11166
Protein SigNET: SLC2A1
Antibody Type: Polyclonal
Antibody Host Species: Rabbit
Antibody Immunogen Source: Synthetic phosphopeptide patterned after human SLC2A1
Antibody Immunogen Sequence: C(βA)QSDK(pT)PEE
Antibody Immunogen Description: Corresponds to amino acid residues Q474 to E481. The effect of T478 phosphorylation is unclear. This is a minor in vivo phosphorylation sites in SLC2A1 based on ≥14 mass spectrometry reports reported in PhosphoSitePlus.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.
Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.
Antibody Concentration: 0.8 mg/ml
Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol
Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.
Product Use: Western blotting | Antibody microarray

Antibody Species Reactivity: This antibody detects the target phosphoprotein in the following species due to conservation of amino acid sequence: Human | Rhesus macaque | Dog | Rat | Mouse | Chicken | Frog.
Scientific Background: SLC2A1 (Solute carrier family 2, facilitated glucose transporter member 1; GLUT1) that belongs to the major facilitator superfamily, Sugar transporter (TC 2.A.1.1) family, Glucose transporter subfamily. It is a crucial transmembrane protein uniporter that facilitates the passive, energy-independent transport of glucose, galactose, and dehydroascorbic acid across the cell plasma membranes. It exhibits a very broad substrate specificity, and can transport a wide range of aldoses including both pentoses and hexoses. It serves as the primary glucose transporter across the blood-brain barrier, red blood cells, and fetal tissues. It can be a cellular receptor for Human T-cell Leukemia Virus (HTLV) I and II. SLC2A1 is found in high concentrations on the plasma membranes of erythrocytes, endothelial cells, and the placenta. Mutations in the SLC2A1 gene cause GLUT1 Deficiency Syndrome (Glut1DS), a metabolic encephalopathy treated with a ketogenic diet. GLUT1 is vital for maintaining cellular metabolic homeostasis and is frequently upregulated in cancer cells to support increased glycolysis.
