Antibody Immunogen Description: Corresponds to amino acid residues C420 to A426. Phosphorylation of Y424 induces interaction with Grb2 and SHP2 PTPN11). This is the major in vivo site for phosphorylation of Shc4 based on ≥44 mass spectrometry reports recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: Shc4 (SHC Adaptor Protein 4; ShcD; RaLP) is a 630-amino acid protein belonging to the Shc family of phospho-tyrosine-binding adapter proteins. It is primarily expressed in the brain, spinal cord, and skeletal muscle, regulating receptor tyrosine kinase (RTK) signaling, neuromuscular junction (NMJ) clustering of acetylcholine receptors, and tumour cell migration/invasion. It functions in anchoring acetylcholine receptors at postsynaptic NMJs, specifically interacting with MuSK (muscle-specific kinase). It plays a role as a modulator of the epidermal growth factor receptor (EGFR) signaling cascade, promoting ligand-independent hyperphosphorylation and regulating receptor trafficking. It contributes to the early phases of agrin-induced tyrosine phosphorylation of CHRNB1. It is found in axons and somata of mature neurons, particularly in the hippocampus, cerebellum, and olfactory bulb. It is also expressed in melanomas and other tumours, where it activates Ras-dependent and -independent migration pathways, contributing to cell proliferation and metastasis. It is highly expressed at the transition from radial growth phase to vertical growth phase and metastatic melanomas, when tumour cells acquire migratory competence and invasive potential.
