Antibody Immunogen Description: Corresponds to amino acid residues R628 to W635. The effect of Y631 phosphorylation is unclear. This is the major site of in vivo phosphorylation of SAC3 based on ≥27 mass spectrometry reports recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: Fig4/Sac3 phosphatase, a 97-kDa protein, specifically removes the 5-phosphate from the inositol ring of PI(3,5)P2, a membrane-bound phospholipid, which acts as a molecular signal for trafficking and fusion of intracellular vesicles and lysosomal function. As part of a complex with PIKfyve and VAC14, it maintains endolysosomal membrane homeostasis. The protein features a SAC-like domain (approx. 400 amino acids) that is essential for its phosphoinositide phosphatase activity, which is conserved across eukaryotes. Besides its lipid activity, FIG4 has serine-protein phosphatase activity that stimulates PIKfyve, contributing to the synthesis of PI(3,5)P2. It is highly expressed in brain and broadly found in all eukaryotic cells and plays a vital role in intracellular membrane trafficking and signalling. Mutations in Fig4/Sac3 have been linked to pale tremor mice that exhibit neuropathy with severe central and peripheral neuronal degeneration, including the loss of neurons in the dorsal root ganglion and the sciatic nerve. Fig4/Sac3 knockout mice have been shown to be smaller in size and exhibit abnormal tremor and gait and neuronal autophagy. Mutations of Fig4 have been linked with Charcot-Marie-Tooth Disease Type 4J (CMT4J), Yunis-Varon Syndrome, Amyotrophic Lateral Sclerosis and Primary Lateral Sclerosis, and Leukoencephalopathy.
