Antibody Immunogen Description: Corresponds to amino acid residues P289 to P296. The effect of T293 phosphorylation is unknown. This is the major phosphosite in NGFR by mass spectrometry (≥6 reports recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: NGFR (Nerve Growth Factor Receptor; p75NTR; CD271, is a 75 kDa transmembrane glycoprotein and member of the tumour necrosis factor receptor (TNFR) superfamily. It binds with low affinity to all neurotrophins—including NGF, BDNF, NT-3, and NT-4/5—regulating neuronal survival, apoptosis, differentiation, and migration. It forms a heterodimeric receptor with SORCS2 that binds the precursor forms of NGF, BDNF and NT-3 with high affinity, and has much lower affinity for mature NGF and BDNF. The receptor consists of an extracellular domain with four cysteine-rich repeats, a single transmembrane domain, and a cytoplasmic death domain. NGFR lacks intrinsic tyrosine kinase activity but functions as a co-receptor with Trk receptors (TrkA, B, C), Sortilin, and LINGO1. It mediates both cell survival and death depending on the cellular context. It is crucial for nerve development, myelination, and plasticity. NGFR plays a role in the inactivation of RHOA, and the regulation of the translocation of GLUT4 to the cell surface in adipocytes and skeletal muscle cells for glucose uptake in response to insulin.
