Antibody Immunogen Description: Corresponds to amino acid residues S780 to P787. The effect of S784 phosphorylation is unknown. This is the major phosphosite in NCAM1 by mass spectrometry (34 reports recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: NCAM1 (Neural cell adhesion molecule 1; CD56) is a multifunctional transmembrane glycoprotein of the immunoglobulin superfamily that mediates neuron-neuron adhesion, neurite fasciculation, outgrowth of neurites, and synaptic plasticity in the nervous system. It is critical for neurodevelopment and immune cell regulation, specifically on NK cells and T lymphocyte subsets. Its adhesive properties are controlled by post-translational modifications, specifically polysialylation. It is also subject to proteolytic cleavage by metalloproteinases like ADAM17 and MMPs. Alternative splicing produces three main isoforms (NCAM-120, -140, -180) that differ in their intracellular domains. The molecule can be post-translationally modified by adding polysialic acid (PSA), which reduces adhesion and promotes cell plasticity. It is expressed on neurons, glial cells, skeletal muscle, and NK cells. It is linked to Alzheimer’s disease due to reduced synaptic levels and implicated in schizophrenia and bipolar disorder.
