Antibody Immunogen Source: Synthetic phosphopeptide patterned after human L1CAM
Antibody Immunogen Sequence: TFGE(pY)RSL(βA)C
Antibody Immunogen Description: Corresponds to amino acid residues T1172 to L1179. The effect of N-CAM L1 Y176 phosphorylation is unclear. This is a major site of in vivo phosphorylation in L1CAM based on ≥10 mass spectrometry reports documented in PhosphoSitePlus. L1CAM is known to be phosphorylated at this site in vitro by Src.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.
Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.

Scientific Background: L1CAM (Neural cell adhesion molecule L1; CD171) is a 200-220 kDa transmembrane glycoprotein from the immunoglobulin superfamily (L1/neurofascin/NgCAM family) that is crucial for nervous system development, including neuronal migration, axon growth and fasciculation, and synapse formation. In the mature brain, plays a role in the dynamics of neuronal structure and function, including synaptic plasticity. It mediates cell-cell adhesion and promotes cell motility. It functions through homophilic (L1-L1) and heterophilic (e.g., L1-integrin) binding. It features an extracellular domain (immunoglobulin-like and fibronectin-type III repeats), a transmembrane segment, and a conserved cytoplasmic domain. Mutations in L1CAM are associated with L1 syndrome (X-linked disorders (CRASH: Corpus callosum hypoplasia, Retardation, Aphasia, Spastic paraplegia, and Hydrocephalus), while its over-expression in various cancers (e.g., ovarian, colon, glioblastoma) drives invasion and chemoresistance. L1CAM is primarily expressed in the central and peripheral nervous systems. This description may include information annotated by UniProt and/or Google AI.
