Antibody Immunogen Description: Corresponds to amino acid residues A361 to Q367. The effect of Y364 phosphorylation is unknown. It is one of the few observed in vivo phosphosites in GABPB1 with ≥1 high throughput mass spectrometry reports recorded in PhosphoSitePlus .
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: GABPB1 (GA-binding protein subunit beta-1) is a transcription factor of the ETS family that interaction with purine rich repeats (GA repeats). GABPB1 acts as the transactivating subunit, combining with GABPA to form a tetrameric complex that that binds DNA to regulate gene expression, including mitochondrial function and cellular growth. It is implicated as a master regulator of nuclear-encoded mitochondrial genes. It is involved in mitochondrial organization and the activation of cytochrome oxidase expression. It is required for expression of the Adenovirus E4 gene. ABPB1 is necessary for the proliferation of neural stem/progenitor cells. GABPB1 supports the activation of the TERT promoter in glioblastoma and other cancers. It plays a role in telomerase activation in cancer cells.
