Antibody Immunogen Description: Corresponds to amino acid residues A299 to D306. The effect of S303 phosphorylation is unclear, but it is the major site of phosphorylation by mass spectrometry (≥14 reports recorded in PhosphoSitePlus).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: GABPA (GA-binding protein alpha chain) is a transcription factor of the ETS family that interacts with DNA with purine rich repeats (GA repeats) such as the PEA3/Ets motif [(C/A)GGA(A/T)(G/A)]. It forms a heterotetrameric complex with GABP to regulate genes involved in mitochondrial function, cell cycle progression, and metabolic energy production. It positively regulates transcription of transcriptional repressor RHIT/ZNF205. It is critically required for normal T-cell development, specifically for the expression of IL-7 receptor chain (IL-7R). It is required for expression of the Adenovirus E4 gene. GABPA plays a complex role in cancer, acting as a tumour suppressor in some contexts (e.g., breast cancer) but being hijacked by TERT promoter mutations in others to drive telomerase expression. GABPA activity is sensitive to cellular redox states, regulated by phosphorylation, and implicated in Down syndrome through its chromosomal location.
