Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: Dvl1 (Segment polarity protein dishevelled homolog DVL-1) is an inhibitor protein that belongs to the Dishevelled family and contains three conserved domains: an N-terminal DIX domain, a central PDZ domain, and a C-terminal DEP domain. It is essential for development, and regulates cell proliferation, differentiation, and neuroblast specification. It binds directly to Frizzled receptors and it plays a role both in canonical and non-canonical Wnt signalling. It participates in Wnt signaling by binding to the cytoplasmic C-terminus of frizzled family members and recruits Axin to the membrane, which inhibits the beta-catenin destruction complex, allowing beta-catenin to accumulate. Its function in the signal transduction pathways mediated by multiple Wnt genes. It is required for LEF1 activation upon WNT1 and WNT3A signalling. DVL1 and PAK1 form a ternary complex with MUSK which is important for MUSK-dependent regulation of AChR clustering during the formation of the neuromuscular junction (NMJ). Dvl1 is typically found in the cytoplasm, it translocates to the plasma membrane upon Wnt activation, and can also be found in the nucleus.
