Antibody Immunogen Description: Corresponds to amino acid residues A781 to L788. The effect of Y784 phosphorylation is unclear, but this is a prominent site phosphorylation of DLG1 by mass spectrometry (≥137 reports recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: Dlg1 (Disks large homolog 1; SAP97) is an essential multidomain adaptor/scaffolding protein of the membrane-associated guanylate kinase (MAGUK) family. It features multiple interaction domains (L27, PDZ, SH3, Hook, and Guanylate kinase domain) that allow it to act as a molecular scaffold/adaptor. It organizes protein complexes at specialized membrane sites to regulate cell polarity, synapse structure, T-cell activation, and epithelial cell-cell junctions. It recruits channels, receptors and signaling molecules to discrete plasma membrane domains in polarized cells. Dlg1 functions in adherens junction assembly, signal transduction, cell proliferation, synaptogenesis and lymphocyte activation. It regulates the excitability of cardiac myocytes by modulating the functional expression of Kv4 channels. As a postsynaptic density (PSD) protein, it interacts with ionotropic glutamate receptors and potassium channels to regulate synaptic plasticity. Dlg1 is a regulator of Kv1.5 channel. It is abundantly expressed in atrial myocardium, and found in lung fibroblasts, cervical epithelial and B-cells. Although it is widely expressed, its diverse isoforms exhibit different expression profiles. It acts as a tumour suppressor by regulating signaling pathways.
