Antibody Immunogen Description: Corresponds to amino acid residues F630 to K637. The effect of T634 phosphorylation is unclear. This is one of the major sites of in vivo phosphorylation of CLTC (≥x1083 reports by mass spectrometry in PhosphoSitePlus). CLTC is known to be phosphorylated at this site in vitro by ZAP70.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: CLTC (Clathrin heavy chain 1) belongs to the clathrin heavy chain family, and is the major 190 kDa protein of the polyhedral coat of coated pits and vesicles. The protein features three heavy chains (1,675 amino acids) and three light chains, forming a 3-legged structure called a triskelion. Two different adapter protein complexes link the clathrin lattice either to the plasma membrane or to the trans-Golgi network. It acts as component of the TACC3/ch-TOG/clathrin complex and is proposed to contribute to stabilization of kinetochore fibers of the mitotic spindle by acting as inter-microtubule bridge. The TACC3/ch-TOG/clathrin complex is required for the maintenance of kinetochore fiber tension. It functions in early autophagosome formation.
