Target Full Name: Nicotinic acetylcholine receptor subunit alpha-9

Target Alias: Nicotinic acetylcholine receptor subunit alpha-9

Product Type Specific: CHRNA9 phosphosite-specific antibody

Antibody Code: PN940

Antibody Target Type: Phosphosite-specific

Antibody Phosphosite: Y430

Protein UniProt: Q9UGM1

Protein SigNET: CHRNA9

Antibody Type: Polyclonal

Antibody Host Species: Rabbit

Antibody Immunogen Source: Synthetic phosphopeptide patterned after human CHRNA9

Antibody Immunogen Sequence: RNIE(pY)IAKC

Antibody Immunogen Description: Corresponds to amino acid residues R426 to C434. The effect of Y430 phosphorylation is unclear, but this is one of the few documented sites of phosphorylation in this protein (≥2 mass spectrometry report recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.

Antibody Concentration: 0.45 mg/ml

Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol

Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.

Product Use: Western blotting | Antibody microarray

Antibody Dilution Recommended: 2 µg/ml for immunoblotting

Antibody Species Reactivity: This antibody detects the target phosphoprotein in the following species due to conservation of amino acid sequence: Human | Dog | Rat | Chicken | Zebra fish.

Scientific Background: CHRNA9 (Neuronal acetylcholine receptor subunit alpha-9) belongs to the ligand-gated ion channel (TC 1.A.9) family, Acetylcholine receptor (TC 1.A.9.1) subfamily, Alpha-9/CHRNA9 sub-subfamily. Agonist binding induces a conformation change that leads to the opening of an ion-conducting channel across the plasma membrane. The channel is permeable to a range of divalent cations including calcium, the influx of which may activate a potassium current which hyperpolarizes the cell membrane. It is primarily located in the outer hair cells (OHCs) of the cochlea, the pituitary gland, keratinocytes, and B- and T-lymphocytes. This Ionotropic receptor may have a role in the modulation of auditory stimuli. In the ear, CHRNA9 stimulation may lead to a reduction in basilar membrane motion, altering the activity of auditory nerve fibers and reducing the range of dynamic hearing. This may protect against acoustic trauma. It may also regulate keratinocyte adhesion. It is involved in tobacco addiction, and polymorphisms are linked to increased risks of non-small cell lung cancer (NSCLC) and tobacco-related breast cancer. CHRNA9-containing receptors are notable for their unique pharmacology, acting more like glycine receptors than typical nicotinic receptors, and appear to play a potential role in mediating neuropathic pain.
