Antibody Immunogen Description: Corresponds to amino acid residues S495 to A502. The effect of T499 phosphorylation is unclear. This is the major in vivo phosphorylation sites in CACNB1 (≥29 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: CACNB1 (Voltage-dependent L-type calcium channel subunit beta-1) is the regulatory subunit of L-type calcium channels and belongs to the calcium channel beta subunit family. It controls the activity of L-type calcium channels that contain CACNA1A as pore-forming subunit. It regulates the activity of L-type calcium channels that contain CACNA1C as pore-forming subunit and increases the presence of the channel complex at the cell membrane. It is required for functional expression L-type calcium channels that contain CACNA1D as pore-forming subunit. It regulates the activity of L-type calcium channels that contain CACNA1B as pore-forming subunit. Isoform 1 and isoform 3 are expressed in brain, heart, spleen, central nervous system and neuroblastoma cells. Isoform 2 is also expressed in skeletal muscle.
