Antibody Immunogen Description: Corresponds to amino acid residues N272 to D279. The effect of S276 phosphorylation is unclear. This is the major in vivo phosphoryation sites with ≥ 128 mass spectrometry reports recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: AP3B1 (Adaptor Related Protein Complex 3 Subunit Beta 1) is a subunit of non-clathrin- and clathrin-associated adaptor protein complex 3 (AP-3) that plays a role in protein sorting in the late-Golgi/trans-Golgi network (TGN) and/or endosomes to lysosomes and lysosome-related organelles (e.g., melanosomes, platelet dense granules). It is involved in melanosome assembly, platelet dense granule organization, and trafficking within neurons, such as anterograde synaptic vesicle transport. The AP complexes mediate both the recruitment of clathrin to membranes and the recognition of sorting signals within the cytosolic tails of transmembrane cargo molecules. AP-3 is also required with the BLOC-1 complex to target cargos into vesicles assembled at cell bodies for delivery into neurites and nerve terminals. It is ubiquitously expressed, with critical roles in immune cells (natural killer and T-cells) and pigment cells. Mutations in AP3B1 lead to HPS2, a rare autosomal recessive disorder characterized by oculocutaneous albinism, bleeding diathesis (due to platelet deficiencies), and immune dysfunction (e.g., neutropenia). AP3B1 is also implicated in the maturation of Weibel-Palade bodies and, when defective, can impair exocytosis through defects in VAMP8 recruitment.
