Antibody Immunogen Description: Corresponds to amino acid residues A692 to E700. Phosphorylation of S696 inhibits receptor desensitization. AKAP12 is known to be phosphorylated at this site in vitro by PKACA .

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: AKAP12 (A-kinase anchoring protein-12) mediates the subcellular compartmentation of protein kinase A (PKA) and protein kinase C (PKC) isoforms as well as Src-family kinases, and calmodulin, enabling crosstalk between these pathways. This adaptor/scaffold protein is expressed in endothelial cells, astrocytes, cultured fibroblasts and osteosarcomas, but not in platelets, leukocytes, monocytic cell lines or peripherical blood cells. It plays a crucial role in maintaining tissue integrity and the blood-brain barrier.It is often silenced in cancers (colon, liver, prostate) through promoter hypermethylation, leading to increased invasion and metastasis. AKAP12 regulates actin-cytoskeleton reorganization, promoting cell flattening and reducing migratory behavior. Loss of AKAP12 leads to hyperactivation of PKC and increased tumour growth, while its regulation is also implicated in neurological diseases like Alzheimer’s and ischemic injury.
