Target Full Name: Vesicle-associated membrane protein-associated protein B/C

Target Alias: ALS8; VAMP (vesicle-associated membrane protein)-associated protein B and C; VAMP-associated protein B/C; VAMP-B; VAMP-C; VAP-B; VAP-C

Product Type Specific: VAPB phosphosite-specific antibody

Antibody Code: PN921

Antibody Target Type: Phosphosite-specific

Antibody Phosphosite: S206

Protein UniProt: O95292

Protein SigNET: VAPB

Antibody Type: Polyclonal

Antibody Host Species: Rabbit

Antibody Immunogen Source: Synthetic phosphopeptide patterned after human VAPB

Antibody Immunogen Sequence: QSN(pS)PISAC

Antibody Immunogen Description: Corresponds to amino acid residues Q203 to A210. The effect of S206 phosphorylation is unclear. This is a minor in vivo site of phosphorylation of VAPB based on ≥12 mass spectrometry reports recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.

Antibody Concentration: 0.6 mg/ml

Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol

Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.

Product Use: Western blotting | Antibody microarray

Antibody Dilution Recommended: 2 µg/ml for immunoblotting

Antibody Species Reactivity: This antibody detects the target phosphoprotein in the following species due to conservation of amino acid sequence: Human | Chimpanzee | Rhesus Macaque | Rat | Mouse | Platypus | Chicken | Frog | Nematode worm | Sea urchin.

Scientific Background: VAPB (Vesicle-associated membrane protein-associated protein B) belongs to the VAMP-associated protein (VAP) (TC 9.B.17) family. It is an endoplasmic reticulm-anchored, tail-anchored protein that is important for membrane trafficking, lipid transport, and the unfolded protein response (UPR). It features an N-terminal Major Sperm Protein (MSP) domain (cytosolic), a coiled-coil domain, and a C-terminal transmembrane anchor. It interacts with FFAT-motif-containing proteins to form membrane contact sites (MCS), bridging the endoplasmic reticulum with other organelles. It is involved in vesicle trafficking, ER-Golgi transport, and initiating the unfolded protein response (UPR). It is Involved in cellular calcium homeostasis regulation. It participates in the endoplasmic reticulum unfolded protein response (UPR) by inducing ERN1/IRE1 activity. VAPB is highly conserved and ubiquitously expressed, often forming homodimers or heterodimers with VAPA. The P56S mutation disrupts normal functioning of VAPB, causing aggregation in the ER, Golgi dispersion, and cell death. Mutations arealso linked to familial Amyotrophic Lateral Sclerosis type 8 (ALS8), a form of motor neuron disease. This description may include information annotated by UniProt and/or Google AI.
