Antibody Immunogen Description: Corresponds to amino acid residues S253 to N2. The effect of T128 phosphorylation is unclear. This is the major in vivo site of phosphorylation of UBQLN2 based on ≥33 mass spectrometry reports recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: UBQLN2 (Ubiquilin 2) functions in the regulation of different protein degradation mechanisms and pathways including ubiquitin-proteasome system (UPS), autophagy and the endoplasmic reticulum-associated protein degradation (ERAD) pathway. It mediates the proteasomal targeting of misfolded or accumulated proteins for degradation by binding (via UBA domain) to their polyubiquitin chains and by interacting (via ubiquitin-like domain) with the subunits of the proteasome. It functions in the ERAD pathway via its interaction with ER-localized proteins FAF2/UBXD8 and HERPUD1 and may form a link between the polyubiquitinated ERAD substrates and the proteasome. It is involved in the regulation of macroautophagy and autophagosome formation in part via conversion of autophagy-related protein LC3 from the cytosolic form LC3-I to the membrane-bound form LC3-II, and it may assist in the maturation of autophagosomes to autolysosomes by mediating autophagosome-lysosome fusion. It negatively regulates the endocytosis of GPCR receptors AVPR2 and ADRB2, by specifically reducing the concentration of receptor-arrestin complexes in clathrin-coated pits.
