Antibody Immunogen Description: Corresponds to amino acid residues M301 to D309. The effects of S304 and S306 phosphorylation are unclear. These are two of the few known phosphorylation sites confirmed by ≥3 and ≥2 mass spectrometry reports recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: STLK6 is a protein-serine/threonine kinase in the STE Group, STE20 family and STLK subfamily. It has been suggested to be a pseudokinase which, in complex with CAB39/MO25 (CAB39/MO25alpha or CAB39L/MO25beta), binds to and activates STK11/LKB1. It adopts a closed conformation typical of active protein kinases and binds STK11/LKB1 as a pseudosubstrate, promoting conformational change of STK11/LKB1 in an active conformation. Ser-184 is present instead of the conserved Asp which is expected to be an active site residue. However, it may be that a phosphorylated Ser-184 might possibly substitute the Asp, although this specific phosphorylation has not been described.
