Antibody Immunogen Description: Corresponds to amino acid residues S266 to S276. The effects of SQSTM1 T269 and S272 phosphorylation are unclear. These are two major sites of in vivo phosphorylation of SQSTM1 (≥208 and ≥335 reports, respectively, from high throughput mass spectrometry studies recorded in PhosphoSitePlus). SQSTM1 is known to be phosphorylated in vitro at T269 and S272 by CDK1 (CDC2), and p38d MAPK (MAPK13).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: SQSTM1 (Sequestome 1 (p62)) is a ubiquitin binding protein that interacts with ubiqutin, providing a scaffold for several signalling proteins and triggering degradation of proteins throught the proteasome or lysosome. It functions as a bridge between polyubiquitinated cargo and autophagosomes. It regulates activation of the nuclear factor kappa-B (NF-kB) signalling pathway. The protein functions as a scaffolding/adaptor protein in concert with TNF receptor-associated factor 6 to mediate activation of NF-kB in response to upstream signals. SQSTM1 also binds autophagosomal membrane protein LC3/ATG8. Mutations in the SQSTM1 gene result in sporadic and familial Paget disease of bone, and have been linked with amyotrophic lateral sclerosis (ALS).
