Antibody Immunogen Description: Corresponds to amino acid residues V104 to H111. The effect of S108 phosphorylation is unclear. This is a major site of in vivo phosphorylation of SOD1 (≥20 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: SOD1 (cytoplasmic copper-zinc superoxide dismutase) serves as an important antioxidant that catalyzes the conversion of the toxic reactive oxygen species superoxide radical O2ˉ anion into oxygen (O2) and hydrogen peroxide (H2O2), which are then metabolized to H2O and O2 by catalase and glutathione peroxidase (1,4). It is an intracellular metalloenzyme that is found in almost every cell in the body (2). In general, SODs play a major role in antioxidant defense mechanisms (3). There are two main types of SOD in mammalian cells. One form (SOD1) contains Cu and Zn ions as a homodimer and exists in the cytoplasm. The two subunits of 16 kDa each are linked by two cysteines forming an intra-subunit disulphide bridge (2). Misfolding of SOD1 has been implicated in amyotrophic lateral sclerosis (ALS). Over 180 different mutations in SOD1 have been linked to ALS. Conformation specific antibodies such as SOD1 (EDI), which targets an exposed region of the dimer interface (EDI) of SOD1, are useful for determining the conformation of SOD1 in affected tissues (5).
