Antibody Immunogen Description: Corresponds to amino acid residues S57 to G63. Y60 phosphorylation induces interaction with ACTA1. This is one of the major in vivo phosphorylation site in profilin 1 (≥68 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: Profilin 1 (PFN1) is a small (~15 kDa), ubiquitously expressed actin-binding protein consisting of 139–140 amino acids. It acts as a key regulator of the actin cytoskeleton by promoting the conversion of actin-ADP to actin-ATP, facilitating polymerization and cell motility. It is also involved in membrane trafficking, signaling, and nuclear processes.At high concentrations, Profilin 1 binds actin and prevents its polymerization, whereas it enhances actin polymerization at low concentrations. In addition to actin, PFN1 binds to poly-L-proline (PLP) motifs on proteins (e.g., VASP, Formins) and phosphoinositides (e.g., PIP2). By binding to PIP2, it inhibits the formation of IP3 and diacylglycerol by sequestering PIP2. It inhibits androgen receptor (AR) and HTT aggregation, and the binding of G-actin that is essential for its inhibition of AR. PFN1 features a globular "profilin-like" fold composed of a seven-stranded beta-sheet flanked by three alpha-helices. Phosphorylation of PFN-1 Tyr-129 induces interaction with VHL. While it is primarily cytosolic, it can be concentrated in dynamic membrane regions.
