Antibody Immunogen Description: Corresponds to amino acid residues R172 to E179. The effect of S176 phosphorylation is unclear. There are no reports of in vivo phosphorylation of this site in KIF5A based on mass spectrometry studies recorded in PhosphoSitePlus.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: KIF5A (Kinesin Family Member 5) belongs to the TRAFAC class myosin-kinesin ATPase superfamily. Kinesin family. Kinesin subfamily. It is a neuron-specific motor protein that acts as an ATP-dependent molecular motor to transport cellular cargo, such as organelles, proteins and RNA granules, along microtubules toward their plus ends (anterograde transport). It is required for slow axonal transport of neurofilament proteins (NFH, NFM and NFL). It consists of an N-terminal motor domain (ATPase activity/microtubule binding), a central coiled-coil stalk (dimerization), and a C-terminal tail (cargo binding). It acts as a dimeric motor, often working in conjunction with Kinesin Light Chains (KLCs). It is distributed throughout the CNS, but it is highly enriched in subsets of neurons.
