Antibody Immunogen Description: Corresponds to amino acid residues G343 to K350. The effect of Y347 phosphorylation is unclear. This is the major in vivo phosphorylation site in HNRNPA1 (≥622 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: hnRNP A1 (Heterogeneous nuclear ribonucleoprotein A1) is a highly abundant, ubiquitously expressed RNA-binding protein. It is primarily localized in the nucleus, but constantly shuttles between the nucleus and cytoplasm to regulate gene expression, often acting as a component of ribonucleosomes. It is involved in alternative splicing, mRNA transport and stability, miRNA processing, and translation regulation. It also plays a role in telomere maintenance and maintaining a contractile vascular smooth muscle cell (VSMC) phenotype. It is composed of two N-terminal RNA-recognition motifs (RRMs) and a C-terminal low-complexity domain (LCD) that facilitates phase separation and stress granule formation. The protein is highly regulated by post-translational modifications (PTMs), which control its subcellular localization and activity in response to stress. Dysfunction of hnRNP A1 is associated with inclusion body myopathy, ALS, and frontotemporal dementia due to its role in cellular stress responses. Mutations in the gene (e.g., in the prion-like domain) are linked to ALS and other neurodegenerative diseases. It acts as a host factor for many viruses (e.g., influenza) to assist in replication, while providing an antiviral effect against others (e.g., HTLV-1).
