Antibody Immunogen Description: Corresponds to amino acid residues A337 to D343. The effect of S340 phosphorylation is unclear. This is a minor in vivo phosphorylation sites in FUS (≥7 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: FUS (Fused in Sarcoma, a DNA-/RNA-binding protein that shares functional homology with TDP-43, which has also been associated with mutations linking to amyotrophic lateral sclerosis (ALS). Also known as hnRNP P2, it plays a role in various cellular processes such as transcription regulation, pre-RNA splicing, RNA transport, DNA repair and damage response. It binds to nascent pre-mRNAs and facilitates RNA polymerase II and U1 small nuclear ribonucleoprotein interaction to couple transcription and splicing. It binds also its own pre-mRNA and autoregulates its expression by non-sense-mediated decay. It shuffles between the nucleus and cytoplasm. It features an N-terminal prion-like domain (involved in aggregation), an RNA recognition motif (RRM), glycine-rich regions, and a C-terminal zinc finger domain and nuclear localization signal (NLS).
