Antibody Immunogen Description: Corresponds to amino acid residues G478 to I485. The effect of Y482 phosphorylation in ANAX11 is unclear. It is the major site of in vivo phosphorylation by mass spectrometry (≥178 reports recorded in PhosphoSitePlus.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ANXA11 (Annexin A11) belongs to the annexin family of calcium- and lipid-binding protein. It is a Ca2+-dependent, 56 kDa phospholipid-binding protein that mediates membrane trafficking and cellular homeostasis. It links membrane dynamics to cell division during mitosis, by translocating from the nucleus to spindle poles in metaphase, to the midzone in anaphase, and to the midbody in telophase. Disruption causes defects in cytokinesis, resulting in cell death. It also acts as a tether between lysosomes and stress granules. Pathological mutations impair this function, leading to the formation of protein aggregates in neurons. It binds specifically to calcyclin in a calcium-dependent manner. It features a conserved C-terminal core with 4 annexin repeats (Ca²⁺-binding) and a unique, long, intrinsically disordered N-terminal domain that acts as a scaffolding site for protein binding (e.g., S100A6/Calcyclin). Mutations in ANXA11, particularly in its N-terminal region, are linked to Amyotrophic Lateral Sclerosis (ALS).
