Antibody Immunogen Description: Corresponds to amino acid residues L488 to L495. The effect of S492 phosphorylation is unclear. This is one of the most frequently phosphorylated in vivo sites in ALS2 from mass spectrometry studies (≥175 reports recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ALS2 (Alsin) may act as a GTPase regulator. It activates Rab5, essential for early endosome trafficking, endocytosis, and endosome-to-lysosome conversion. ALS2 facilitates endosomal trafficking of growth factor receptors, protecting against cytotoxicity and maintaining neuronal stability. It controls survival and growth of spinal motoneurons. ALS2 contains three key predicted GEF domains: an N-terminal RCC1-like domain, a central Dbl homology and pleckstrin homology (DH/PH) domain, and a C-terminal Vacuolar Protein Sorting 9 (VPS9) domain. Three alternatively spliced human isoforms are known. The protein is found in the cytoplasm, membranes, and centrosomes, where it associates with Rab5-positive early endosomal compartments. Mutations in the ALS2 gene cause recessive neurodegenerative disorders, including juvenile lateral sclerosis and infantile-onset ascending spastic paralysis. The absence of alsin leads to impaired endosome-to-lysosome conversion and abnormal accumulation of trafficking-related vesicles in motor neurons, causing retrograde degeneration.
