Antibody Immunogen Description: Corresponds to amino acid residues R720 to K730. Phosphorylation of T724 and S726 induces activation. ADD1 is known to be phosphorylated in vitro at T724 by CDK5, and at S726 by Chk1, PKACa (PRKACA), PKCa (PRKCA), PKCd (PRKCD), and PKCz (PRKCZ).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ADD1 (adducin-1, adducin-alpha) is a membrane-cytoskeleton-associated protein that binds calmodulin and promotes the assembly of the spectrin-actin network. It belongs to the aldolase class II family, Adducin subfamily. It acts as a heterodimer to cap actin filaments and recruit spectrin to the actin-spectrin network, crucial for membrane stability. It consists of a protease-resistant globular head, a connecting subdomain, and a protease-sensitive tail region. It functions primarily as a heterodimer with beta- or gamma-adducin. It regulates cell shape, adhesion, and signaling, and is implicated in human hypertension and cardiovascular disease through genetic polymorphisms. It is ubiquitously expressed in all tissues, and it is found in much higher levels in reticulocytes than the adducin beta subunit.
